The base is known for serving as the home base of two Boeing VC-25 aircraft which have the call sign Air Force One while the President of the United States is on board. As of the 2010 census, the resident population was 2,973. The host at Andrews is the 316th Wing, assigned to the Air Force District of Washington. Andrews was best known for its special air mission role, the transportation of senior government and military leaders. President Harry S. Truman was the first to fly a presidential flight out of Andrews on 24 November 1946. In 1958, when airspace around Naval Air Station Anacostia in Washington, D. C became too crowded, naval air activities were moved to Andrews Air Force Base to facilitate jet operations with a detachment of T-2V SeaStar jet trainers. In September 2007, NAF Washington was disestablished and was merged with the reserve center at NAF, creating the largest NOSC in the country. The most significant realignment to Andrews AFB was the relocation of Naval Air Facility Washington by the Realignment and Closure Commission in May 2005. By October 2006, the Navy Operational Support Center, Naval District Washington Reserve Command, assumed responsibility for the transport of VIPs and light cargo for the Navy and Marine Corps. The Naval Air Reserve Command was dis established in September 2007 and was merging with the Maryland Adelphi Reserve Command in September 2008 to form the National Air Force Reserve Command (NAFRC).

In September 2009, the NAFRC was disestablished and merged with NAFWashington Reserve Command to create NAFAdelphi. In October 2010, the National Security Agency (NSA) announced that the NSA would no longer be responsible for transporting VIPs or light cargo to and from Andrews AFB. In December 2010, NSA announced that it would be consolidating all of its operations at the NSA’s National Air and Space Operations Center (NASOC) in Maryland. NASOC became the new headquarters for the NSA in September 2011. In March 2012, the NSA announced it was consolidating its operations in Maryland and moving its operations to the NSA’s National Security Operations Center in Virginia. In April 2013, the Pentagon announced that NASC would become the new home of the National Defense Authorization Act for Fiscal Year 2014 (NADAA). In March 2014, the FAA announced that its new headquarters in Virginia would also be the home of NADSOC. In January 2015, NASCO announced that they would be moving their operations to Virginia. The move was the largest realignment of the country in the history of the military. In May 2015, the U.S. Air Force announced it would relocate its operations from Virginia to Maryland. In July 2016, the base was the site of a joint operation between the NSA and the Department of Defense.
